Alexander Plein erlangte sein Diplom zum Bühnendarsteller an der Stage School in Hamburg. Bereits während des Abschlussjahres ging er mit dem Nimmerland Musical »Peter Pan« in der Rolle des Michael auf Deutschlandtour. Zu Beginn seiner Karriere führte es den gebürtigen Düsseldorfer zu den Burgfestspielen Jagsthausen. Dort wurde er als Teil des Ensembles im Musical »Catch Me If You Can« mit dem Publikumspreis ausgezeichnet. Seither verkörperte er den Romeo in der Lahnsteiner Inszenierung von »Romeo und Julia« sowie Adson von Melk in »Der Name der Rose« bei den Lahnsteiner Burgspielen. Er tourte im Ensemble von »My Fair Lady«, »Alice im Wunderland«, »Der kleine Prinz«, »Chaplin - Das Musical« und mit der Arena Show der beliebten TV-Serie »Paw Patrol Live«. Weiterhin war er in der 70er Jahre Schlager -Komödie »Immer wieder sonntags« in Lübeck und Hanau, »The Addams Family« in Merzig, als Diva in der gleichnamigen Travestie-Revue in Malentes Theaterpalast in Bonn und als Sänger im »Palazzo Colombino Basel« engagiert und brachte das Publikum des Scala Theaters in Köln in der musikalischen Komödie »Drei Prumme För Pitter« zum Lachen. Am Schlosspark Theater Berlin war er in »Monty Python's Spamalot« zu sehen. Alexander tourte mit den Kölner Symphonikern in »Die Künstler-WG« und »Broadway Dreams«. Mit »Harry Potter und das verwunschene Kind« im Mehr! Theater am Großmarkt zog es ihn zurück nach Hamburg, wo er im Anschluss sein Debut an Schmidts Tivoli auf der Reeperbahn in »Die Weihnachtsbäckerei Das Musical« feierte. Als Swing spielte er die Erfolgsproduktion mit den Liedern von Rolf Zuckowski ebenso im Theater des Westens Berlin, im Metropol Theater Bremen und im Theater am Marientor Duisburg.
Im Sommer 2025 ist Alexander erstmals bei den Burgfestspielen Bad Vilbel in »Hallo, Dolly!« als Barnaby Tucker und »West Side Story« als Big Deal engagiert. Im Tonstudio leiht Alexander sowohl realen als auch animierten Charakteren seine Stimme. Ebenso ist er Sprecher für Werbung, Imagefilme, Hörbücher und Hörspiele, Apps und Spiele sowie E-Learning. Seit 2023 ist er in den Guinness World Records gelistet. Als einer von 623 Sprechern vertonte er das Buch „In jeder Träne spiegelt sich die Sonne“ von Iris Ferraldi. Zuletzt stand Alexander für die TV-Serien NOTRUF und Auf Streife - Die neuen Einsätze (Sat.1 / Joyn) vor der Kamera. Im Webinar In nur 2h zum Profi-Presenter erklärt er seine Tipps und Tricks zum Vortragen und Sprechen vor Menschen. Für Hapag-Lloyd Cruises war Alexander mit der MS Europa als Tänzer auf dem Indischen Ozean unterwegs. Er choreographierte Kampagnen von Beiersdorf NIVEA und war Moderator zahlreicher Galas unter anderem für den LIONS CLUB. Ebenso ist Alexander Teil der Werbespots von KUBOTA, Tempo, En Vogue, Trimet und Mercedes Me. He was also engaged in the 70s hit comedy »Sundays again and again« in Lübeck and Hanau, »The Addams Family« in Merzig, as a Diva in the same named drag revue show at Malentes Theaterpalast in Bonn and as a singer at »Palazzo Colombino Basel« and made the audience at the Scala Theater in Cologne laugh in the musical comedy »Drei Prumme För Pitter«.
